| Grid preparation | |
|---|---|
| User actions to be taken in advance of visiting eBIC for microscope sessions | |
| Optimise grids | grids should be well optimised in advance |
| Pre-screen grids | pre-screen prior to visiting - eBIC does not have resources to offer on-site sample preparation |
| Select grids | only ship grids you know you want to collect high-resolution data from |
| Sufficient grids | send a sufficient number of pre-screened grids to fill the full allocation of instrument time |
| Clip grids (Krios sessions) |
if users have access to an FEI auto-grid clipping station, eBIC expect users to clip in advance if users do not have access to a clipping station, eBIC will clip a max of 4 grids per 48-hours |
|
Number of grids |
bring no more than 4 pre-screened grids per sample a total of 11 grids can be loaded per 48 or 72-hour session sessions longer than 72-hours can have a second load grids can be recovered upon request, minus the one in the column |
| cryoFIB grids |
for cryoFIB sessions only 2 grids will be loaded per session |
| Sample description | |
|---|---|
| Users will need to provide the information below in advance of visiting eBIC for microscope sessions | |
| Proposal information |
all samples must be associated to and described in a successful BAG or rapid proposal all proposals require evidence of suitable pre-screened grids (acceptance criteria) the BAG or rapid proposal number should be provided with your sample |
| User Administration System (UAS) |
principal investigators will be prompted to submit sample information and experimental team into the UAS - once submitted the principal investigator will receive a confirmation email containing the experimental details |
| Experimental risk assessment (ERA) |
enter all samples that users are bringing to Diamond, eBIC into the ERA samples not on the ERA will NOT be loaded into microscopes |
| Hazard Group 2 or biological specimens |
samples requiring decontamination with agents other than 70% ethanol can not be handled at eBIC if a user's HG2 sample can be decontaminated with ethanol this must be evidenced in the ERA |
