Overview 

This is a hybrid event targeted at researchers new to the field. The course will provide fundamental lectures, demonstrations and practicals in cellular sample preparation, FIB milling, lamella preparation, cryo-ET data collection, initial quality assessment and data processing.

Lectures will be held at Diamond Light Source and will be broadcast to the wider community via Zoom. Demonstrations and hands-on practical sessions covering cellular sample preparation, correlative FIB milling, lamella preparation, cryo-ET data collection, initial quality assessment and data processing will be available to in-person attendees only.

Programme

Day 1: Monday 8th September 2025

  • Lectures covering an introduction to cryo-ET workflows, cellular sample preparation and sample vitrification (in-person and virtual)
  • Demonstration covering sample vitrification
  • Practical covering cryo-ET sample preparation and sample vitrification (in-person only)

Day 2: Tuesday 9th September 2025

  • Demonstration of cryo-ET screening and data collection (in-person and virtual)
  • Practical covering cryo-ET screening and data collection (in-person only)

Day 3: Wednesday 10th September 2025

  • Lectures covering correlative cryo-ET workflows, cryo-FIB-SEM lamella preparation and Soft X-ray Tomography (in-person and virtual)
  • Practicals covering principles of cryo-FIB-SEM set up and introduction to lamella preparation (in-person only)

Day 4: Thursday 11th September 2025

  • Practicals covering principles of cryo-FIB-SEM set up and introduction to lamella preparation (in-person only)

Day 5: Friday 12th September 2025

  • Lectures covering cryo-ET image processing and reconstruction (in-person and virtual)
  • Practical sessions covering cryoET image processing (in-person only) 
  • Student learning and feedback discussion (in-person only)
