Three-dimensional imaging at the water window using soft X-rays is becoming increasingly relevant in answering biologically and environmentally relevant questions. The compatibility of soft X-ray tomography with other imaging methods such as electron microscopy, super-resolution fluorescence microscopy, X-ray fluorescence and X-ray absorption spectroscopy, makes it even more appealing to the scientific community who rely on high-resolution imaging datasets to extract detailed information aimed at solving real world problems.
However, the fully automated alignment of different angular projections required to generate a tomographic X-ray volume as well as the 3D correlation of X-ray tomograms with 3D datasets from fluorescence microscopy remains a bottleneck. This is because, within the three-dimensional space of cells, there are no features to track continuously.
Preliminary data have been collected at the beamline on the internalisation of an array of nanoparticles, however, this summer studentship project aims to take it a step further by functionalising these nanoparticles for improved cellular uptake. Apart from the obvious gains of alignment and correlation of datasets that we aim to derive from these nanoparticles, they will be trialled as cargos for the delivery of drugs and antigens to their target locations within cell populations.
