Load Pucks Directly

The simplest way to load samples into Unipucks is to freeze the sample directly into the puck at the home laboratory. Remember to wear the correct personal protective equipment when working with liquid nitrogen.

  • Cool the main puck in liquid nitrogen.
  • When you have the crystal as required in the loop, simply place it directly into the puck.
  • When all 16 positions are full, or all the crystals have been frozen, cool the base plate of the puck.
  • Align the base plate with the puck and push firmly in to place.
  • Store the puck in liquid nitrogen until the synchrotron visit.

Load Vial Pucks

If you do not have pucks in your home laboratory, pucks can be loaded from frozen crystals stored in vials. Remember to wear the correct personal protective equipment when working with liquid nitrogen.

  • Cool the vial puck in liquid nitrogen.
  • Transfer your frozen samples, in vials, into the vial puck.
  • When all 16 positions are full, or all the crystals have been transferred, cool the base plate of the puck.
  • Align the base plate with the puck and push firmly in to place.
  • Maintain the vial puck in liquid nitrogen to load the robot.
